A novel method for early detection of MIC value – Broth dilution using indicator solution versus agar dilution: an original article

چکیده

Introduction: The adequate protocol for treatment of an infection is often determined on the basis minimum inhibitory concentration (MIC) causative organism. Traditional methods (agar dilution, microbroth and gradient diffusion) are labour intensive time consuming (they usually take over 48 hours to report results). On other hand, automated systems (VITEK™, Phoenix™, MicroScan WalkAway™) rapid MIC detection (using dielectrophoresis (DEP), magnetic bead rotation sensors microfluidic incubation) require expensive instruments. This study aimed develop a method with ability applied resource limited setting. Methods: Agar dilution novel broth (containing indicator solution) were simultaneously performed using amikacin, ceftriaxone, piperacillin-tazobactam, imipenem, cefoxitin azithromycin. Results: Isolates Escherichia coli, Enterobacter spp, Klebsiella Staphylococcus aureus, Proteus mirabilis, Acinetobacter spp Pseudomonas used. values Enterobacteriaceae S. aureus isolates each antibiotic obtained within 4 5 by method. corresponded shown following day agar Conclusion: Broth solution effective in determination cephalosporins, penicillin, carbapenems, cephamycin, aminoglycosides macrolides most aureus. Unfortunately this did not work non-fermenter group organisms like as their results could be before 24 hours. saving, relatively inexpensive applicable settings.

Comparison of agar dilution, tube dilution, and broth microdilution susceptibility tests for determination of ramoplanin MICs.

Standard broth microdilution (with and without bovine serum albumin [BSA] supplementation), tube dilution, and agar dilution susceptibility tests were compared for determining ramoplanin MICs. With a data base of 246 clinical isolates of gram-positive bacteria from 33 U.S. sites, it was shown that (i) agar and tube dilution susceptibility tests gave essentially the same results (93.9% of the te...

Comparison of broth microdilution method using Haemophilus test medium and agar dilution method for susceptibility testing of Eikenella corrodens.

Susceptibility testing of Eikenella corrodens is usually performed by a Mueller-Hinton sheep blood agar dilution (AD) method. However, this method is impractical for testing only a few strains. We compared AD with the broth microdilution method using Haemophilus test medium (HTM) in order to determine the susceptibility of 36 clinical isolates of E. corrodens to eight antimicrobial agents. MICs...

Detection with Agar Dilution Susceptibility Testing

A dye reduction method for determining a definitive endpoint with agar dilution susceptibility testing has been developed. Bacterial growth was determined by applying to the inoculum spot a dye solution containing 2-(p-iodophenyl)-3-(pnitrophenyl)-5-phenyltetrazolium chloride and phenazine methosulfate. Viable colonies reduced the tetrazolium salt to a visible red color within 3 to 5 min. The m...

Comparison of the agar dilution, tube dilution, and broth microdilution susceptibility tests for determination of teicoplanin MICs.

The purpose of this study was to evaluate the National Committee for Clinical Laboratory Standards agar dilution, tube dilution, and broth microdilution susceptibility tests for the measurement of teicoplanin MICs. The three standardized tests gave equivalent (within a twofold dilution) results with 98.8 to 99.0% of the 508 gram-positive clinical isolates tested, indicating that either method m...

A broth dilution method of assaying streptothricin and streptomycin.

The agar dilution (Waksman, 1943) and the agar diffusion (Foster and Woodruff, 1943b) methods of assay of streptothricin and streptomycin are the procedures most commonly employed and have led to what Waksman calls "dilution units" (Waksman, 1943) and "diffusion units" (Schatz, Bugie, and Waksman, 1944). However, in addition, Waksman speaks of "Escherichia coli units," or "Bacillus subtilis uni...
